Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Structurally, Melanotan-2 retains the core recognition motif of alpha-melanocyte-stimulating hormone while adding a lactam bridge that links two side chains and constrains the molecule into a ring. This modification lowers susceptibility to enzymatic degradation. The compound acts as an agonist at melanocortin receptors, particularly subtypes associated with melanin production. Because the same receptor family influences several physiological processes, researchers note that its activity is not confined to pigmentation alone. Receptor selectivity continues to be examined in published studies.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its sequence incorporates modified residues that increase potency and extend biological activity relative to the native hormone. The compound binds receptors of the melanocortin family and is examined mainly in laboratory research. It does not occur naturally and exists only as a manufactured chemical entity produced by solid-phase synthesis.

Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Melanotan II is a synthetic cyclic heptapeptide analog derived from the core sequence of alpha-melanocyte-stimulating hormone. Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. The compound is not an approved medicine in any major jurisdiction and appears mainly in laboratory and research-chemical settings. Its structure incorporates a lactam bridge between side chains, which constrains the ring and slows enzymatic breakdown relative to the natural hormone.
Unlike the related medication eplerenone, spironolactone is said to not be metabolized by CYP3A4. However, hepatic CYP3A4 is likely responsible for the 6β-hydroxylation of 7α-TMS into 6β-OH-7α-TMS. 7α-TMS may also be hydroxylated at the C3α and C3β positions. Spironolactone is dethioacetylated into canrenone. Finally, the C17 γ-lactone ring of spironolactone is hydrolyzed by the paraoxonase PON3. It was originally thought to be hydrolyzed by PON1, but this was due to contamination with PON3.
The Ukrainian Air Force claimed that twenty-five missiles were fired at Kyiv and other parts of Ukraine. Twenty-three were intercepted by air defence. While there were no reports of casualties, there was some property damage. Ukraine also said it had moved its Victory Day celebrations to May 8 in line with the rest of Europe, while 9 May will be called Europe Day. French journalist Arman Soldin, who worked for Agence France Presse, was killed in a rocket attack in Chasiv Yar, near Bakhmut. He was with a team of AFP journalists travelling with Ukrainian soldiers when the group came under fire from Grad rockets.
=== Assumed regenerative models === Stem cells are thought to mediate repair via five primary mechanisms: 1) providing an anti-inflammatory effect, 2) homing to damaged tissues and recruiting other cells, such as endothelial progenitor cells, that are necessary for tissue growth, 3) supporting tissue remodeling over scar formation, 4) inhibiting apoptosis, and 5) differentiating into bone, cartilage, tendon, and ligament tissue. To further enrich blood supply to the damaged areas, and consequently promote tissue regeneration, platelet-rich plasma could be used in conjunction with stem cell transplantation. The efficacy of some stem cell populations may also be affected by the method of delivery; for instance, to regenerate bone, stem cells are often introduced in a scaffold where they produce the minerals necessary for generation of functional bone. Stem cells have been shown to have low immunogenicity due to the relatively low number of MHC molecules found on their surface. In addition, they have been found to secrete chemokines that alter the immune response and promote tolerance of the new tissue. This allows for allogeneic treatments to be performed without a high rejection risk.
The vast majority of reported cases of foodborne illness occur as individual or sporadic cases. The origin of most sporadic cases is undetermined. In the United States, where people eat outside the home frequently, 58% of cases originate from commercial food facilities (2004 FoodNet data). An outbreak is defined as occurring when two or more people experience similar illness after consuming food from a common source. Often, a combination of events contributes to an outbreak, for example, food might be left at room temperature for many hours, allowing bacteria to multiply which is compounded by inadequate cooking which results in a failure to kill the dangerously elevated bacterial levels. Outbreaks are usually identified when those affected know each other. Outbreaks can also be identified by public health staff when there are unexpected increases in laboratory results for certain strains of bacteria. Outbreak detection and investigation in the United States is primarily handled by local health jurisdictions and is inconsistent from district to district. It is estimated that 1–2% of outbreaks are detected. When an outbreak is suspected, investigators conduct an epidemiologic assessment, laboratory analysis, and environmental assessment simultaneously. Investigators construct an epidemic curve, a histogram of cases over time, to characterize transmission patterns and narrow the exposure window. Case-control studies are commonly used, comparing foods eaten by ill individuals against those eaten by healthy controls to identify statistically significant exposures.
=== Faith and beliefs === When asked if he believed in God, Freeman said: "It's a hard question because as I said at the start, I think we invented God. So if I believe in God, and I do, it's because I think I'm God." He later said that his experience working on The Story of God with Morgan Freeman did not change his views on religion, further stating that he sees Zoroastrianism being a belief system, that, "is intrinsically me: 'Good thoughts, good words, good deeds' sums it up. How many of us that believe in that?".

Using the co-ordinates of the subunits and segments to determine the topographic location of the defect on the nose, the plastic surgeon plans, maps, and executes a rhinoplasty procedure. The unitary division of the nasal topography permits minimal, but precise, cutting, and maximal corrective-tissue coverage, to produce a functional nose of proportionate size, contour, and appearance for the patient. Hence, if more than 50 percent of an aesthetic subunit is lost (damaged, defective, destroyed) the surgeon replaces the entire aesthetic segment, usually with a regional tissue graft, harvested from either the face or the head, or with a tissue graft harvested from elsewhere on the patient's body.
=== United Fruit Co. and United Brands Company subsidiary === In 1963, the company was sold again, followed by another sale in 1967 to the United Fruit Co. conglomerate. AMK Corporation purchased United Fruit in 1970. Then AMK formed United Brands Company to hold A&W. In 1971, A&W Beverages Inc.—a beverage subsidiary—began supplying bottled A&W products to grocery stores. The bottled products would become available nationally. In 1972, A&W's Canadian division was sold to Unilever. A&W attempted to open restaurants in mainland Japan in the early 1970s, specifically in Fukuoka prefecture and the regions of Kanto and Kansai. The chain's performance on the mainland was sluggish in contrast to Okinawa due to several factors, such as the 1973 oil crisis, prompting the chain to withdraw from the market. A&W's Japanese operations are still handled from Okinawa. There were further fruitless attempts to bring the chain to the mainland: in the 1980s, the Okinawan branch briefly attempted to open restaurants in Kagoshima Prefecture but ended up limiting itself to Okinawa, while in the 2000s a pilot restaurant existed in Tokyo, which was quickly withdrawn. 1978 saw the introduction of a standard menu for use in all restaurants in the USA. In the 1970s, A&W had more stores than McDonald's, with a peak in 1974 of 2,400 units.
Dyson says that the report showed that, even from a narrow military point of view, the US was better off not using nuclear weapons. Dyson opposed the Vietnam War and the invasion of Iraq. He supported Barack Obama in the 2008 US presidential election and The New York Times described him as a political liberal. He was one of 29 leading US scientists who wrote Obama a strongly supportive letter about his administration's 2015 nuclear deal with Iran.
=== Anxiety === Benzoctamine's main clinical use is for the treatment of anxiety, and evidence points to it being as effective as other clinical anxiety drugs, in particular diazepam. In the treatment of symptoms of mild anxiety due to psychoneurosis, a daily dosage of 30 to 80 mg of benzoctamine was shown to be just as effective as 6–20 mg of diazepam. In another study one group of patients were given 10 mg of benzoctamine three times a day, while another group was given 5 mg of diazepam, and the treatments were equivalent. While these studies point to higher doses of benzoctamine being needed to exert the same pharmacological effects, the drug is still popular because of its ability to act as an anxiolytic without producing the common respiratory depression associated with other sedative drugs. Some studies have even shown that it stimulates the respiratory system.

== Crimes == Pressdee intentionally administered lethal doses of insulin to patients across five nursing homes in Pennsylvania. The first two victims she was charged with murdering were James Bartoe, 55, and Joseph Campbell, 83. They died on December 4 and 25, 2022, respectively. Her third victim was Nicholas Cymbol, 43, who died on May 1, 2023. Pressdee told her attorney that she believed she was ending the suffering of her victims.
The use of concrete in construction grew rapidly from 1850 onward, and was soon the dominant use for cements. Thus Portland cement began its predominant role. Isaac Charles Johnson further refined the production of meso-Portland cement (middle stage of development) and claimed he was the real father of Portland cement. Setting time and "early strength" are important characteristics of cements. Hydraulic limes, "natural" cements, and "artificial" cements all rely on their belite (2 CaO · SiO2, abbreviated as C2S) content for strength development. Belite develops strength slowly. Because they were burned at temperatures below 1,250 °C (2,280 °F), they contained no alite (3 CaO · SiO2, abbreviated as C3S), which is responsible for early strength in modern cements. The first cement to consistently contain alite was made by William Aspdin in the early 1840s: This was what we call today "modern" Portland cement. Because of the air of mystery with which William Aspdin surrounded his product, others (e.g., Vicat and Johnson) have claimed precedence in this invention, but recent analysis of both his concrete and raw cement have shown that William Aspdin's product made at Northfleet Cement Works, Kent was a true alite-based cement. However, Aspdin's methods were "rule-of-thumb": Vicat is responsible for establishing the chemical basis of these cements, and Johnson established the importance of sintering the mix in the kiln.
=== Ministerial changes === The first ministerial change occurred in July 2023. Daniela Carneiro, after requesting disaffiliation from União Brasil, was replaced by Celso Sabino (UNIÃO-PA) in the Ministry of Tourism. On 6 September 2023, the first ministerial reform of the government was carried out, with the aim of obtaining greater governability. Lula replaced Ana Moser with the deputy André Fufuca (PP-BA) in the Ministry of Sport and Márcio França (PSB-SP) with Silvio Costa Filho, of the Republicanos, in the Ministry of Ports and Airports. Márcio França assumed the newly created Ministry of Entrepreneurship and Small Businesses. On Wednesday, 31 January 2024, Lula officially dismissed Flávio Dino from the leadership of the Ministry of Justice, after having nominated him to the position of justice of the Supreme Federal Court (STF) in the seat left by Rosa Weber with her compulsory retirement. To replace him, Ricardo Lewandowski was appointed, a former member of the Supreme Court who had left the body in the previous year, also due to compulsory retirement. On 6 September 2024, the Minister of Human Rights, Silvio Almeida, was dismissed from the position after it was disclosed that the NGO MeToo Brasil had received complaints of sexual harassment against him. On 9 September, the president announced that the Minas Gerais State deputy Macaé Evaristo (PT) will occupy the leadership of the Ministry of Human Rights.
=== Belief === Morea was raised in a Roman Catholic household and has openly celebrated Christian festivals such as Easter with his family. In 2023, Morea was invited among several members of India's Christian community to a Christmas celebration hosted by Prime Minister Narendra Modi at his official residence in New Delhi.
About 1,500 heat-related deaths occurred in the Netherlands, again largely the elderly. The heat wave broke no records, although four tropical weather-designated days in mid-July, preceding the official wave, are not counted due to a cool day in between and the nature of the Netherlands specification/definition of a heat wave. The highest temperature recorded this heatwave was on 7 August, when in Arcen, in Limburg, a temperature of 37.8 °C (100.0 °F) was reached, 0.8 °C below the national record (since 1904). A higher temperature had only been recorded twice before. On 8 August, a temperature of 37.7 °C (99.9 °F) was recorded, and 12 August had a temperature of 37.2 °C (99.0 °F). Initially, 141 deaths were attributed to the heat wave in Spain. A further research of INE estimated a 12,963 excess of deaths during summer of 2003. Temperature records were broken in various cities, with the heat wave being more felt in typically cooler northern Spain. Record temperatures were felt in:

No. It is a synthetic analogue carrying four amino acid changes, a lactam ring and an amidated C-terminus. The natural hormone is a linear thirteen-amino-acid peptide processed from proopiomelanocortin.
Afamelanotide is the linear analogue [Nle4-D-Phe7]-alpha-MSH, sometimes called melanotan I, while melanotan II is cyclic and carries three further substitutions. The two are distinct molecules and are not interchangeable in analytical testing.
Binding to the four melanocortin receptor subtypes is well documented in vitro. The relative contribution of each subtype to whole-body effects in humans is far less certain.
It is a synthetic cyclic heptapeptide and an analogue of alpha-melanocyte-stimulating hormone. The molecule is produced by chemical synthesis rather than extracted from a biological source.
