Regulatory treatment varies by country. In the United States the peptide is not approved as a medicine, and products offered for human use may be treated as unapproved new drugs; some states also restrict sale. Australia, the United Kingdom, and European Union member states apply comparable restrictions to unapproved peptide products. Border agencies have seized shipments labelled as research chemicals. Classification may change over time, and the legal position for personal importation is not clearly settled in most published guidance.
Lyophilised melanotan-2 is supplied as a solid, which is more stable than a solution. The material is hygroscopic, so weighing is done quickly, in low humidity, with the container kept sealed. Reconstitution usually uses water for injection or bacteriostatic water, added down the wall of the vial to limit foaming. A reconstituted solution is held at 2 to 8 °C and kept away from light. Repeated freezing and thawing of the same vial is avoided because ice crystal formation and concentration effects degrade the peptide.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.

Regulatory status differs by country, and in many places supplying the compound for human consumption is unlawful. Vendors frequently label material as intended for research use only, a designation that shifts stated purpose but does not create a legal pathway for personal use. Certificates of analysis accompanying such products vary widely in detail and provenance. Third-party testing exists but is voluntary, and results are rarely linked to a specific lot in a publicly verifiable way.
Identity testing for a synthetic peptide relies on several complementary methods. Reversed-phase high-performance liquid chromatography separates the target from related impurities and reports purity as a percentage of total peak area. Mass spectrometry confirms molecular mass and can reveal deletions or truncations. Amino acid analysis and peptide mapping provide sequence-level confirmation, while counter-ion content and residual solvents are measured separately. A purity figure alone does not establish identity, so a complete dataset combines chromatographic and spectrometric evidence.
Lyophilized peptide powder is generally stored frozen, protected from light and moisture. Tryptophan residues are susceptible to oxidation, and the lactam bridge can hydrolyze under strongly acidic or basic conditions. Solutions prepared for laboratory work degrade faster than dry powder, and repeated freeze-thaw cycles accelerate loss. Common practice is to aliquot solutions before freezing and to avoid alkaline buffers. Reported stability windows vary with concentration, buffer, and temperature, so exact shelf lives are method-specific rather than universal.
Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Melanotan II is a synthetic cyclic heptapeptide analog derived from the core sequence of alpha-melanocyte-stimulating hormone. Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. The compound is not an approved medicine in any major jurisdiction and appears mainly in laboratory and research-chemical settings. Its structure incorporates a lactam bridge between side chains, which constrains the ring and slows enzymatic breakdown relative to the natural hormone.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.
Solid peptide kept dry at minus twenty degrees Celsius, shielded from light and moisture, is generally considered stable for extended periods. Solutions are divided into single-use aliquots and held at minus twenty or minus eighty degrees Celsius, because repeated freeze-thaw cycles promote aggregation and loss of material to container surfaces. Hydrolysis of the backbone and oxidation of tryptophan are the principal degradation routes in aqueous solution, and both accelerate at ambient temperature. Hygroscopic uptake after a vial is opened can also shift the actual mass weighed, which affects any concentration calculated from it.
Receptor studies place melanotan-2 among non-selective melanocortin agonists, binding MC1R, MC3R, MC4R and MC5R rather than a single subtype. Activation of MC1R on cutaneous melanocytes raises tyrosinase activity and shifts pigment synthesis toward eumelanin, which is darker and more photostable than pheomelanin. Central receptors, particularly MC4R, are associated with appetite suppression and with reported effects on sexual function. Because subtype selectivity is low, the same molecule engages pigment, metabolic and vascular pathways at once, and this breadth is a common explanation offered for the range of adverse events described in user reports.
No regulatory authority has approved melanotan-2 for human use, and several countries classify it as a prescription-only or controlled substance, which restricts lawful supply. Material sold online is generally labelled as a research chemical and is not required to meet pharmaceutical standards of identity or purity. Published human data consist mainly of small uncontrolled studies, case reports and adverse-event notifications, so the evidence base is descriptive rather than confirmatory. Whether repeated melanocyte stimulation alters long-term naevus behaviour remains an open question that no completed trial has resolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous tridecapeptide that regulates pigment production. Two modifications distinguish it from the natural hormone: norleucine replaces methionine at the N-terminus, which limits oxidation, and a D-phenylalanine substitution raises receptor affinity. The ring is closed through an aspartate-lysine lactam bridge, giving the molecule a constrained conformation. The free base has a molecular mass near 1024 daltons, and commercial material is usually supplied as an acetate salt. It appears in the literature as a research peptide rather than an approved therapeutic agent.
The Bergmann degradation is intended for and has been used as a method for peptide sequencing. It was also proposed for use in cleaving the 3,4-bond of the penicillin nucleus. The compound 2,2-dimethyl-6-phthalimido-3-penamyl isocyanate was arrived at through various means, including the Curtius rearrangement, and it was envisioned that it could undergo the Bergmann degradation to form the desired aldehyde as well as the urea by-product. Though the Bergmann degradation was indeed possible, it was discovered that simple dilute acid hydrolysis would suffice in forming the desired product.
No paralogs of c7orf26 have been found in the human genome, however, six unique isoforms have been identified. They are c7orf26 isoform (X1, X2, X3, X4) and isoform 2 (two sub-isoforms identified). Below is a table of a variety of orthologs of the human c7orf26. The table include closely, intermediately and distantly related orthologs. Orthologs of the human protein c7orf26 are listed above in descending order of the date of divergence. c7orf26 is highly conserved throughout all orthologs, this is demonstrated with a 65% identity in the least similar ortholog. c7orf26 has evolved slowly and evenly over time. Below is a phylogenetic tree showing the evolutionary history of c7orf26 and its nearest orthologs.
== Silicon-28 == Silicon-28, the most abundant isotope of silicon, is of particular interest in the construction of quantum computers when highly enriched, as the presence of 29Si in a sample of silicon contributes to quantum decoherence. Extremely pure (>99.9998%) samples of 28Si can be produced through selective ionization and deposition of 28Si from silane gas. Due to the extremely high purity that can be obtained in this manner, the Avogadro project sought to develop a new definition of the kilogram by making a 93.75 mm (3.691 in) sphere of the isotope and determining the exact number of atoms in the sample. Silicon-28 is produced in stars during the alpha process and the oxygen-burning process, and drives the silicon-burning process in massive stars shortly before they go supernova.
Miglitol is an oral alpha-glucosidase inhibitor used in the treatment of type 2 diabetes. It works by reversibly inhibiting alpha-glucosidase enzymes in the small intestine, which delays the digestion of complex carbohydrates and subsequently reduces postprandial glucose levels. Approved for clinical use since 1998, miglitol has demonstrated efficacy in improving glycemic control, reducing HbA1c levels, and decreasing both fasting and postprandial plasma glucose concentrations in long-term clinical trials. Additionally, recent studies have suggested that miglitol may have potential as an anti-obesity agent, showing promise in reducing body weight and body mass index in obese or diabetic patients. While generally well-tolerated, the most common side effects associated with miglitol are gastrointestinal disturbances, which are typically mild to moderate and tend to decrease over time. It must be taken at the start of main meals to have maximal effect In contrast to acarbose (another alpha-glucosidase inhibitor), miglitol is systemically absorbed; however, it is not metabolized and is excreted by the kidneys.
=== Location === Catecholamines are produced mainly by the chromaffin cells of the adrenal medulla and the postganglionic fibers of the sympathetic nervous system. Dopamine, which acts as a neurotransmitter in the central nervous system, is largely produced in neuronal cell bodies in two areas of the brainstem: the ventral tegmental area and the substantia nigra, the latter of which contains neuromelanin-pigmented neurons. The similarly neuromelanin-pigmented cell bodies of the locus coeruleus produce norepinephrine. Epinephrine is produced in small groups of neurons in the human brain which express its synthesizing enzyme, phenylethanolamine N-methyltransferase; these neurons project from a nucleus that is adjacent (ventrolateral) to the area postrema and from a nucleus in the dorsal region of the solitary tract.

=== History === The team first competed in the Victorian Junior Football League (later the AFL reserves) when the competition was established in 1919, and was known as Essendon Juniors until the end of 1922. The team enjoyed success in the form of eight premierships, including in the last year of the reserves competition in 1999. From 2000 until 2002, the club's reserves team competed in the Victorian Football League. At the end of 2002, the club dissolved its reserves team and established a reserves affiliation with the Bendigo Football Club in the VFL. The affiliation ran for ten years from 2003 until 2012, allowing reserves players from the Essendon list to play with Bendigo. For all but the final year of the affiliation, Bendigo was known as the Bendigo Bombers. The club re-established its reserves team in 2013, seeking greater developmental autonomy. The team plays its home games at Windy Hill. The team is made up of senior-listed AFL players and VFL-contracted players.
=== Routes for topical drug administration === In general, there are three possible routes for drug administration into or across the skin. The first one is the transcellular route, where the drugs are required to pass through both lipid matrix and dead corneocytes of the stratum corneum. The second one is the intercellular route, where the drugs only have to pass through lipid domains between corneocytes. The third one is the trans-appendageal route, where the drugs are transported by hair follicles, sebaceous glands, or sweat glands. The most common route for drug delivery into the skin is the intercellular route. Multiple steps are involved.
Life is thought to have emerged from inanimate matter more than 3.5 billion years ago when a rudimentary abiogenesis process gradually evolved into an autocatalytic process capable of template-based replication. It was proposed on the basis of experimentally feasible RNA reactions catalyzed by a ribozyme, that the emergence of life was likely a gradual process involving the evolutionary properties of variation, heredity and reproduction, ultimately allowing for Darwinian evolution. Recent efforts have been directed at trying to demonstrate RNA replication under conditions that assume the presence during early evolution of plausible nucleotide intermediates and plausible environmental conditions that could favor strand replication alternating with strand separation. One such effort was the demonstration of high fidelity RNA copying using 2',3'-cyclic phosphate ligation to allow polynucleotide synthesis under conditions also compatible with strand separation. In another study, it was shown that in a model oscillating Hadean environment likely to have been abundant during early evolution, that ribozyme-mediated RNA synthesis and replication can occur. Catalysis The ability to catalyze simple chemical reactions—which would enhance creation of molecules that are building blocks of RNA molecules (i.e., a strand of RNA that would make creating more strands of RNA easier). Relatively short RNA molecules with such abilities have been artificially formed in the lab. A recent study showed that almost any nucleic acid can evolve into a catalytic sequence under appropriate selection.
=== Animal feed === Amino acids are sometimes added to animal feed because some of the components of these feeds, such as soybeans, have low levels of some of the essential amino acids, especially of lysine, methionine, threonine, and tryptophan. Likewise amino acids are used to chelate metal cations in order to improve the absorption of minerals from feed supplements.

Enthesitis, wherein there is inflammation of the entheses, the sites where tendons or ligaments insert into the bone. It is associated with HLA B27 arthropathies such as ankylosing spondylitis, psoriatic arthritis, and reactive arthritis. Apophysitis, inflammation of the bony attachment, generally associated with overuse among growing children.
=== Penicillin G === Penicillin G is destroyed by stomach acid, so it cannot be taken by mouth, but doses as high as 2.4 g can be given (much higher than penicillin V). It is given by intravenous or intramuscular injection. It can be formulated as an insoluble salt, and there are two such formulations in current use: procaine penicillin and benzathine benzylpenicillin. When a high concentration in the blood must be maintained, penicillin G must be administered at relatively frequent intervals, because it is eliminated quite rapidly from the bloodstream by the kidney. Penicillin G is licensed for use to treat septicaemia, empyema, pneumonia, pericarditis, endocarditis and meningitis caused by susceptible strains of staphylococci and streptococci. It is also licensed for the treatment of anthrax, actinomycosis, cervicofacial disease, thoracic and abdominal disease, clostridial infections, botulism, gas gangrene (with accompanying debridement and/or surgery as indicated), tetanus (as an adjunctive therapy to human tetanus immune globulin), diphtheria (as an adjunctive therapy to antitoxin and for the prevention of the carrier state), erysipelothrix endocarditis, fusospirochetosis (severe infections of the oropharynx, lower respiratory tract and genital area), Listeria infections, meningitis, endocarditis, Pasteurella infections including bacteraemia and meningitis, Haverhill fever; rat-bite fever and disseminated gonococcal infections, meningococcal meningitis and/or septicaemia caused by penicillin-susceptible organisms and syphilis.
Both trials enrolled participants 18 to 75 years old with moderate to severe glabellar lines. Participants received a single intramuscular injection of daxibotulinumtoxinA or placebo at five sites within the muscles between the eyebrows. The most common side effects of daxibotulinumtoxinA are headache, drooping eyelids, and weakness of facial muscles. LetibotulinumtoxinA (Letybo) was approved for medical use in the United States in February 2024. It is indicated to temporarily improve the appearance of moderate-to-severe glabellar lines. The FDA approved letibotulinumtoxinA based on evidence from three clinical trials (BLESS I [NCT02677298], BLESS II [NCT02677805], and BLESS III [NCT03985982]) of 1,271 participants with moderate to severe wrinkles between the eyebrows for efficacy and safety assessment. These trials were conducted at 31 sites in the United States and the European Union. All three trials enrolled participants 18 to 75 years old with moderate to severe glabellar lines (wrinkles between the eyebrows). Participants received a single intramuscular injection of letibotulinumtoxinA or placebo at five sites within the muscles between the eyebrows. The most common side effects of letibotulinumtoxinA are headache, drooping of eyelid and brow, and twitching of eyelid.
2025: Tristan Tan Yi Rui, who was originally condemned to death for trafficking not less than 337.6 grams of methamphetamine, later had his sentence commuted to life imprisonment after President Tharman Shanmugaratnam granted his clemency plea. This marked the first successful clemency plea in 27 years.

Low temperature slows hydrolysis and oxidation, the two main routes by which the peptide backbone and side chains are modified. A lyophilised powder stored at −20 °C is more stable than one kept at room temperature, and once the material is dissolved the degradation rate rises, making refrigeration more important.
Liquid chromatography coupled to mass spectrometry is the most common approach because it combines a retention time with a mass measurement. A chromatographic peak alone cannot establish which peptide is present, so mass determination or amino acid analysis is used as an orthogonal confirmation alongside the separation.
This depends on buffer, pH, and concentration, and published stability data for this peptide are limited. In practice laboratories work through a refrigerated solution within a few weeks and discard samples that show cloudiness or visible particles. Freezing and thawing repeatedly is generally discouraged.
No. It is a laboratory-made peptide, while the natural hormone is alpha-melanocyte-stimulating hormone, a longer peptide produced by the pituitary gland and by skin cells. Melanotan-2 mimics only a short active region of that hormone and contains non-natural residues such as D-phenylalanine.
