Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Reversed-phase high-performance liquid chromatography is the routine method for purity assessment. Peptides absorb near 214 nm because of the peptide bond, and a gradient of acetonitrile in water separates the intact peptide from deletion sequences, oxidised products, and earlier-eluting fragments at neutral pH. Electrospray ionisation mass spectrometry provides an orthogonal check: the measured mass must agree with the theoretical value. Amino acid analysis and peptide mapping confirm structure but are used less often. Reference standards remain scarce because the peptide is not described in any pharmacopoeia.
Regulatory treatment varies by country. In the United States the peptide is not approved as a medicine, and products offered for human use may be treated as unapproved new drugs; some states also restrict sale. Australia, the United Kingdom, and European Union member states apply comparable restrictions to unapproved peptide products. Border agencies have seized shipments labelled as research chemicals. Classification may change over time, and the legal position for personal importation is not clearly settled in most published guidance.
Lyophilised melanotan-2 is supplied as a solid, which is more stable than a solution. The material is hygroscopic, so weighing is done quickly, in low humidity, with the container kept sealed. Reconstitution usually uses water for injection or bacteriostatic water, added down the wall of the vial to limit foaming. A reconstituted solution is held at 2 to 8 °C and kept away from light. Repeated freezing and thawing of the same vial is avoided because ice crystal formation and concentration effects degrade the peptide.

No regulatory authority has approved melanotan-2 for human use, and several countries classify it as a prescription-only or controlled substance, which restricts lawful supply. Material sold online is generally labelled as a research chemical and is not required to meet pharmaceutical standards of identity or purity. Published human data consist mainly of small uncontrolled studies, case reports and adverse-event notifications, so the evidence base is descriptive rather than confirmatory. Whether repeated melanocyte stimulation alters long-term naevus behaviour remains an open question that no completed trial has resolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous tridecapeptide that regulates pigment production. Two modifications distinguish it from the natural hormone: norleucine replaces methionine at the N-terminus, which limits oxidation, and a D-phenylalanine substitution raises receptor affinity. The ring is closed through an aspartate-lysine lactam bridge, giving the molecule a constrained conformation. The free base has a molecular mass near 1024 daltons, and commercial material is usually supplied as an acetate salt. It appears in the literature as a research peptide rather than an approved therapeutic agent.
Receptor studies place melanotan-2 among non-selective melanocortin agonists, binding MC1R, MC3R, MC4R and MC5R rather than a single subtype. Activation of MC1R on cutaneous melanocytes raises tyrosinase activity and shifts pigment synthesis toward eumelanin, which is darker and more photostable than pheomelanin. Central receptors, particularly MC4R, are associated with appetite suppression and with reported effects on sexual function. Because subtype selectivity is low, the same molecule engages pigment, metabolic and vascular pathways at once, and this breadth is a common explanation offered for the range of adverse events described in user reports.
Verification of a purchased sample requires documentation linking a batch to a certificate of analysis, and that document should be read for the methods used rather than the headline purity figure. A single chromatographic percentage does not establish identity. Independent laboratories can perform identity and content assays, but no such test establishes that a product is suitable for human use. Claims about efficacy rest largely on small, early studies rather than on replicated controlled trials, and that gap remains open.
Identity testing for a cyclic peptide of this size usually relies on reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The mass spectrum confirms molecular weight, while the chromatographic trace indicates the proportion of related impurities. Tandem mass spectrometry can provide sequence-level information when fragmentation data are compared against a reference standard. Nuclear magnetic resonance is sometimes used to confirm the lactam bridge, although it requires more material and greater operator expertise than routine chromatographic methods.
== Specificity == Specificity is conferred by the fact that, of the four bonds of the alpha-carbon of the amino acid aldimine state, the bond perpendicular to the pyridine ring will be broken (Dunathan Stereoelectronic Hypothesis). Consequently, specificity is dictated by how the enzymes bind their substrates. An additional role in specificity is played by the ease of protonation of the pyridine ring nitrogen.
== Applications == Linear elasticity is used widely in the design and analysis of structures such as beams, plates and shells, and sandwich composites. This theory is also the basis of much of fracture mechanics. Hyperelasticity is primarily used to determine the response of elastomer-based objects such as gaskets and of biological materials such as soft tissues and cell membranes.
=== Bacteriophage Morphology === In 1956, Kaesberg and his colleagues studied staphylococcus bacteriophages via electron microscopy and found that freeze-dried particles and air-dried particles had significant morphological differences, specifically in size and shape. They were then able to classify the phages into two serological categories: large with flattened heads and long tails and smaller with long tails and circular heads. They noted that phages within the same class had structural similarities, and phages from different classes differed in structure.
However, light intensity walking is an effective way to help control casual glucose level (CGL), but a minimum of 6000 steps must be achieved daily to have consistent effectiveness in controlling CGL.Nevertheless, there is no significant correlation between light intensity walking and HbA1c, therefore regular moderate intensity exercise is advised, specifically aerobic exercise has been proven to improve both fasting and postprandial blood glucose, insulin dosage, and insulin usage within the body. It is still contested which form of exercise/physical activity is best for pregnant women, yet some movement is better than no movement. Although exercise does not reduce the risk of developing GDM, it does help reduce some of the associated risks. When it comes to exercise in pregnant women who have GDM there is a decrease in the risk of having a newborn with macrosomia, decrease in maternal weight gain and a decrease in c-sections. Although exercise is not the cure for GDM, it does help pregnant women decrease any complications or risk factors that can arise from the disease. Self-monitoring can be accomplished using a handheld capillary glucose dosage system. Compliance with these glucometer systems can be low. There is not a lot of research into what target blood sugar levels should be for women with gestational diabetes, and targets recommended to women vary globally. Target ranges advised by the Australasian Diabetes in Pregnancy Society are as follows:

==== Remoting ==== Component methods may be made available as web services with no additional coding and configuration. All that is required is for a method's access to be declared 'remote'. ColdFusion automatically generates a WSDL at the URL for the component in this manner: http://path/to/components/Component.cfc?wsdl. Aside from SOAP, the services are offered in Flash Remoting binary format. Methods which are declared remote may also be invoked via an HTTP GET or POST request. Consider the GET request as shown.
acronym = an abbreviation pronounced as if it were a word, e.g., SARS = severe acute respiratory syndrome, pronounced to rhyme with cars initialism = an abbreviation pronounced wholly or partly using the names of its constituent letters, e.g., CD = compact disc, pronounced cee dee pseudo-blend = an abbreviation whose extra or omitted letters mean that it cannot stand as a true acronym, initialism, or portmanteau (a word formed by combining two or more words). (a) = acronym, e.g.: SARS – (a) severe acute respiratory syndrome (i) = initialism, e.g.: CD – (i) compact disc (p) = pseudo-blend, e.g.: UNIFEM – (p) United Nations Development Fund for Women (s) = symbol (none of the above, representing and pronounced as something else; for example: MHz – megahertz) Some terms are spoken as either acronym or initialism, e.g., VoIP, pronounced both as voyp and V-O-I-P. Any joint, including the temporomandibular joints, cervical spine, hips, and tiny joints of the hands and feet, may be impacted. Myalgia and tenosynovitis are two other typical musculoskeletal symptoms. SJIA is characterized by splenic and lymph node enlargements, which can happen separately or simultaneously. Particularly prevalent locations for prominent symmetrical lymphadenopathy are the anterior cervical, axillary, and inguinal regions. The enlarged lymph nodes are usually firm, movable, and nontender. Splenomegaly, which affects less than 10% of individuals, typically manifests within the first few years of onset and can be severe. Pericardial involvement frequently occurs. In one study, 81% of children with active systemic symptoms had abnormal echocardiographic findings, and 36% of patients had an effusion or pericardial thickening. The majority of pericardial effusions don't cause any symptoms; they can only be identified by echocardiography and don't show any overt cardiomegaly or usual ECG abnormalities. The characteristic sign of acute pericarditis is chest pain, especially when laying supine and with or without dyspnea. The most frequent respiratory symptom, pleural effusions, can be associated with pericarditis. They are typically asymptomatic and only discovered by chance on chest radiography.
=== Financial markets === Thai equities rallied when trading resumed on 9 February. The SET Index closed at 1,400.89, up 46.88 points or 3.46 per cent, having reacted at the open to unofficial results that pointed to Bhumjaithai leading the next government and easing fears of a political vacuum. Turnover reached 102.1 billion baht, the first hundred-billion-baht session in a year and five months. Foreign investors bought a net 16.5 billion baht and institutions a net 6.2 billion, while domestic retail investors sold a net 24.3 billion. Therdsak Thaveeteeratham of Asia Plus Securities called the rally a surprise, saying the market read the result as improving the odds of a stable government formation and of sustained fund flows back into Thai stocks. Asadej Kongsiri, president of the Stock Exchange of Thailand, said investors expected continuity from the outgoing administration and welcomed policies aimed at domestic demand and living costs. Several analysts warned that profit-taking was likely after so sharp a jump.
ACS first established technical divisions in 1908 to foster the exchange of information among scientists who work in particular fields of chemistry or professional interests. Divisional activities include organizing technical sessions at ACS meetings, publishing books and resources, administering awards and lectureships, and conducting other events. The original five divisions were 1) organic chemistry, 2) industrial chemists and chemical engineers, 3) agricultural and food chemistry, 4) fertilizer chemistry, and 5) physical and inorganic chemistry. As of 2016, there are 32 technical divisions of ACS.

=== Nuclear isomers, including a "stable" one === The 251 known stable nuclides include tantalum-180m, since even though its decay is automatically implied by it being "metastable", this has not been observed. All "stable" isotopes (stable by observation, not theory) are the ground states of nuclei, except for tantalum-180m, which is a nuclear isomer or excited state. The ground state, tantalum-180, is radioactive with half-life 8 hours; in contrast, the decay of the nuclear isomer is extremely strongly forbidden by spin-parity selection rules. It has been reported by direct observation that the half-life of 180mTa to gamma decay must be >1015 years. Other possible modes of 180mTa decay (beta decay, electron capture, and alpha decay) have also never been observed.
== Effect == This receptor has an inhibitory function on most of the tissues in which it rests. In the brain, it slows metabolic activity by a combination of actions. At the neuron's synapse, it reduces synaptic vesicle release.
=== Maturana, narco-fútbol and the return to the World Cup (1987–1989) === The transformation began in May 1987, when the federation appointed Francisco Maturana — a former Once Caldas and Atlético Nacional coach then in charge of the youth setup — to the senior side. Maturana discarded the reactive schemes of his predecessors for a possession-based approach built on short passing, zonal marking and a crowded midfield, and licensed René Higuita to operate far from his goal as a sweeper-keeper — a style Colombian supporters came to call toque-toque.
== Contraindications == Artesunate is typically a well tolerated medicine. Known contraindications include a previous severe allergic reaction to artesunate. Drugs that should be avoided while on artesunate are the drugs that inhibit the liver enzyme CYP2A6. These drugs include amiodarone, desipramine, isoniazid, ketoconazole, letrozole, methoxsalen and tranylcypromine.
Here, the resulting dimer has a stoichiometry different from the initial pair of monomers. Disaccharides need not be composed of the same monosaccharides to be considered dimers. An example is sucrose, a dimer of fructose and glucose, which follows the same reaction equation as presented above. Amino acids can also form dimers, which are called dipeptides. An example is glycylglycine, consisting of two glycine molecules joined by a peptide bond. Other examples include aspartame and carnosine.

It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
No national medicines agency has approved melanotan-2 for clinical use. Afamelanotide, a related but distinct peptide, holds a marketing authorisation in the European Union for a rare photosensitivity disorder. Melanotan-2 itself is handled as a laboratory chemical.
Its broad activity across melanocortin receptors makes it a tool for probing pigmentation, appetite and vascular signalling. Early trials recorded skin darkening and other effects that were not the original focus of the work. Those observations generated hypotheses that later studies have examined.
Low temperature slows hydrolysis and oxidation, the two main routes by which the peptide backbone and side chains are modified. A lyophilised powder stored at −20 °C is more stable than one kept at room temperature, and once the material is dissolved the degradation rate rises, making refrigeration more important.
